بررسی ایمنیزایی واکسن پلیتوپ کاندید HIV مبتنی بر DNA در مدل موشی و تاثیر رویکردهای استفاده از ادجوانت آلوم و تزریق زیرجلدی بر میزان کارآیی آن
Investigation of Immunization of DNA-Based Polyepitop HIV Vaccine Candidate in Mouse Model and the Impact of Alum Adjuvant and Subcutaneous Infusion on its Efficiency
<div style="text-align: justify;"><span style="font-size:12px;"><span style="font-family:iransharp;"><strong>اهداف:</strong> یکی از چالشهای دنیای امروز و یکی از اولویتهای بهداشت جهانی، مبارزه با همهگیری بیماری ایدز <span dir="LTR">(AIDS)</span> است. مطالعات نشان داده است که دامنه و وسعت القای پاسخهای ایمنی در برابر <span dir="LTR">HIV</span> و القای همزمان پاسخهای ایمنی خونی و سلولی در افزایش کارآیی واکسنهای کاندید <span dir="LTR">HIV</span> بسیار تاثیر گذار است. از این رو، اخیراً رویکردهایی نظیر استفاده از واکسنهای پلیاپیتوپی و افزودن ادجوانتها در فرمولاسیون واکسنهای کاندید علیه <span dir="LTR">HIV</span> بسیار مورد توجه قرار گرفته است.<br>
<strong>مواد و روشها:</strong> در این تحقیق، پس از ترانسفورماسیون و تکثیر وکتور بیانی یوکاریوتی <span dir="LTR">pcDNA۳.۱-tat/pol/gag/env</span> در میزبان پروکاریوتی <em><span dir="LTR">E. coli</span></em><span dir="LTR"> (DH۵α)</span>، استخراج وکتور و آمادهسازی آن انجام شد. سپس واکسنهای کاندید <span dir="LTR">DNA</span>ای بهصورت زیرجلدی، در روزهای صفر، ۱۴ و ۲۸ به موشهای ماده رده <span dir="LTR">BALB/c</span> تزریق شده و نهایتاً پاسخهای ایمنی خونی و سلولی القاشده توسط آنها ارزیابی شد.<br>
<strong>یافتهها:</strong> نتایج مطالعه نشان داد که واکسنهای <span dir="LTR">DNA</span>ای مذکور با روش تزریق زیرجلدی بهخوبی قادر به القای پاسخهای ایمنی (سلولی و خونی) نبودند.<br>
<strong>نتیجهگیری:</strong> این مشاهده میتواند بهدلیل نقص در هر یک از مراحل برداشت وکتور توسط سلول هدف تا بیان و ارایه سطحی اپیتوپها از یک طرف، یا ناکارآمدی روش تزریق زیرجلدی از طرف دیگر باشد. از این رو، ممکن است سایر روشهای تزریق یا رهایش واکسن بههمراه استفاده از ادجوانتهای مختلف در فرمولاسیون واکسن کاندید، بتوانند پاسخهای ایمنی بهتری را القا کنند.</span></span></div>
<strong>Aims:</strong> One of the challenges of today's world and also global health priorities is pandemicity of AIDS. Studies have shown that the scope and breadth of the immune responses induction are very effective to protect against HIV. Moreover, simultaneous induction of humoral and cellular immunity responses increases the effectiveness of candidate HIV vaccines. Hence, new approaches such as polyepitopic vaccine strategy and addition of different adjuvants in HIV vaccines’ formulations have been recently considered.<br>
<strong>Materials and Methods:</strong> In the present study, eukaryotic expression vector (pcDNA3.1-<em>tat</em>/<em>pol</em>/<em>gag</em>/<em>env</em>) was transformed and amplified in the prokaryotic host cells <em>E. coli</em> (DH5α). After vector extraction, it was concentrated and formulated alone and in combination with Alum adjuvant and used as DNA candidate vaccines. DNA candidate vaccines were, then, subcutaneously injected to the BALB/c mice on 0, 14, and 28 days and elicited humoral and cellular immunity responses were finally evaluated.<br>
<strong>Findings:</strong> The results showed that the candidate DNA vaccine could not efficiently induce immunity responses (both humoral and cellular responses) by subcutaneous route injection.<br>
<strong>Conclusion:</strong> This observation can be due to a defect in each of the steps of vector harvesting by the target cell to express the surface presentation of the epitopes on the one hand, or the inefficiency of the subcutaneous injection method on the other. Therefore, other vaccines’ injection and deliveries routes along with addition of other adjuvants in vaccine’s formulations could induce immunity responses efficiently and increase vaccine efficacy.
