جداسازی وشناسایی باکتری‌های تولیدکننده ی α- آمیلاز همزیست درختان حرا از سواحل غربی جزیره قشم (کنار سیاه) و بررسی برخی از خصوصیات بیوشیمیایی α- آمیلاز در باکتری های Bacillus sp. HR10 و Bacillus sp. HR11

چکیده
هدف از مطالعه حاضر جداسازی و شناسایی باکتری های بومی تولید کنند آنزیم آلفا آمیلاز همزیست با درختان حرا در جزیره قشم ، هرمزگان ، ایران بود. در این مطالعه 45 باکتری از ریشه و برگ Avicennia marina از سواحل غربی جزیره قشم (کنارسیاه) جداسازی شد. از میان آنها 18 سویه توانایی تولید آلفا آمیلاز را داشتند. آنالیز توالی ژن 16S rRNA مربوط به سویه های دارای بیشترین تولید آنزیم نشان داد که هر دو سویه جداسازی شده متعلق به جنس باسیلوس شامل Bacillus sp.HR10 و Bacillus sp.HR11 می باشند. بر اساس نتایج تاثیر دما و pH بر روی تولید آنزیم نشان داده شد که سویه های HR10و HR11 دارای بیشترین تولید آنزیم به ترتیب در دمای 35 و 30 درجه سانتی گراد و 8 pH می باشند. نتایج تعیین ویژگی بیوشیمیایی آنزیم نشان داد که سویه های HR10 و HR11 دارای بیشترین فعالیت آمیلازی در دمای 70 و 60 درجه سانتی گراد و pH بهینه 8 می باشند. نتایج این مطالعه نشان می دهد که باکتری های همزیست جدا شده از A. marina منبع بالقوه ای از آنزیم آلفا آمیلاز بوده که دامنه گسترده ای از دما و pH را متحمل می شوند. بطور کلی براساس نتایج حاصل، این سویه های باکتریایی می توانند منبع مناسبی جهت تولید آنزیم آمیلاز متحمل به حرارت با پایداری عملکردی بالا جهت کاربرد در صنعت باشند.
