Abstract

Physalis alkekengi L. is planted in gardens and green spaces because of the beautiful and colorful sepals surrounding the fruit. The species is widely used in traditional medicine and treating a range of diseases. Micropropagation of P. alkekengi was evaluated using the node and internode explants. After sterilization and seed germination, sterile seedlings were transformed to basal MS medium to create sterilized seedlings as a source of explants. Regeneration of nodes and internodes explants was studied at various concentrations of growth regulators of 2, 4-D and BAP as well as in medium lacking growth regulators or control (11 various media). The experiment was conducted in a completely randomized design with three replicates. The internodal explants produced shoot on media 2 (0.2mgl-12, 4-D), 3 (0.2mgl-12, 4-D+0/2mgl-1 BAP ), and 4 (0.5mgl-12, 4-D+0.2mgl-1 BAP ) and then were rooted on these media. The nodal explants in control and different hormonal treatments generated shoots; interestingly, shoots generated in control medium successfully established roots on the same medium after 7 days (70%). The other regenerated shoots in different media (10) were rooted on ½ MS medium containing 1mgl-1IBA. The rooted plants were transplanted into pots containing sand as well as perlite to be well acclimated before transfer to the greenhouse. They grew well later in the greenhouse at a 100% success This study shows high in vitro regeneration capability of this species as an important medicinal and ornamental plant. Therefore, it is suggested to use this species in molecular and genetic studies, somaclonal variation, and the production of herbal medicine.

Abstract

Flowering transition is one of the most important developmental processes of higher plants, which is controlled by endogenous and external environmental signals. These signaling cues are perceived in leaves and shoot apical meristem (SAM) to induce flower formation. APETALA1 (AP1) is one of floral meristem identity genes that regulate the specification and formation of floral meristems and is required for sepals and petals formation. In this study, the expression of this gene in different organs of Eruca sativa as well as the effect of brassinosteroids (BRs) on flowering and the gene expression was investigated. RNA was extracted from different organs and first-strand cDNA was synthesized. Specific primers were designed based on the sequence alignment of AP1 isoform genes from other plants. In the vegetative stage, no expression was observed in different organs. Brassinosteroid treatment from 28 days (vegetative stage) to flower buds formation caused early flowering, so that the treated plants flower about 10 days earlier than the control. In addition, plant size and its organs were larger in plants treated with brassinosteroids. Evaluation of EvsAP1 gene expression in reproductive phase showed its expression in flower buds, sepals and petals but no was seen in roots, stems, leaves, stamen and gynocium. Also, the onset of expression of this gene was observed earlier, indicating that the transition to flowering and flower bud formation occurs faster in treated plants; therefore, expression occurs earlier. However, expression levels did not affected by brassinosteroids and no significant difference was observed between treated and control samples.

Abstract

Wave conditions have a significant effect on the hydrodynamic behavior of OWC. As the interactions of the WEC and the incident waves are important, therefore, due to the importance of coefficients in evaluating the performance of the OWC, in this paper, the experimental evaluation of dimensionless hydrodynamic coefficients of a MC-OWC is applied. To define the experimental tests, considering the installation location of the converter on the break water, the conditions of the Caspian Sea implemented. Calibration and uncertainty analysis have performed, experimental tests have been carried out in the wave tank of the BNUT. According to the results, assuming a dimensionless water depth, with increasing dimensionless frequency of the wave, the dimensionless coefficient of transmitted wave, the dimensionless coefficient of reflected wave, dimensionless coefficient of discharge and the dimensionless coefficient of pressure increase. The results showed that due to the change of dimensionless wave number from 1.9 to 3.3, discharge coefficients, reflected wave, pressure and transmitted wave are 1.6 times, 2.2 times, 2.8 times, respectively, are 3.5 times, the dimensionless coefficient of the transmitted wave is highly sensitive to the wave conditions; the dimensional coefficient of discharge will have less changes compared to other coefficients. On the other hand, the results showed that the OWC in this study has an efficiency of 41.8% in the best case. This efficiency occurs at the dimensionless natural frequency of 0.88 and the dimensionless water intake depth of 0.032; under these conditions, the amplitude of water fluctuations inside the OWC reaches 9.6 cm.

Abstract

AbstractPeptides obtained from protein hydrolysis have many bioactive properties and have strong antioxidant activity against free radicals and prevent oxidation processes that cause damage to biological macromolecules and degradation and loss of food quality. Due to the large amount of rice production in the world, a large amount of rice bran is produced and available. As a suitable and cheap protein source, rice bran can be used for the production of plant-derived peptides. Optimizing conditions of enzymatic hydrolysis of rice bran protein by alcalase enzyme was done with the aim of achieving maximum antioxidant properties. In order to check the antioxidant activity of the obtained peptides, iron 3 reduction power tests and DPPH free radical inhibitory activity and total antioxidant activity were used. In order to optimize the process, Design Expert software and response surface method were used with three independent variables: enzyme concentration to substrate 1-3%, temperature 40-55 degrees Celsius and hydrolysis time 30-210 minutes. The optimal treatment was obtained under the determined conditions including temperature of 51.5 degrees Celsius, time of 131.5 minutes and concentration of enzyme to substrate of 3%, which has maximum DPPH free radical inhibition of 37.172%, total antioxidant activity of 1.109% and reductiveness of 2.084%. The obtained results showed that the hydrolysis process of rice bran protein by alkalase enzyme has led to the production of peptides with high and significant antioxidant properties that can be used in the production of useful foods and pharmaceutical industries and can also replace synthetic antioxidants.

Abstract

With increasing intercultural communication among people from diverse linguistic and cultural backgrounds, the integration of language and culture has recently been foregrounded in language education. This has led to a growing need for studies on intercultural communicative competence. To address this need, this study aimed to afford an insight into the beliefs of 20 Iranian language teachers on intercultural language teaching (ICLT). Based on a mixed-method design, two different instruments were used to collect the data. First, an ICLT questionnaire which included 12 items was administered to 20 teachers. This study also used semi-structured interviews to obtain an in-depth understanding of the EFL teachers’ attitudes toward ICLT. The findings from the questionnaire revealed that the teachers were favorably disposed toward ICLT and thought that culture teaching is as important as language teaching. They believed that language teachers are required to inform the learners of intercultural issues. Also, the thematic content analysis of the interviews led to the discovery of four main themes, namely definition of culture, ICLT and its significance, challenges in ICLT, and sources used by the teachers to improve their intercultural knowledge. However, many teachers’ responses in the interviews indicated some degree of uncertainty regarding the concept of ICLT, as they were not confident how to integrate language and culture or how to deal with challenges in implementation of ICLT. These findings imply as the teachers’ perceptions would guide their practices in the classroom, more attention needs to be devoted to the inclusion of ICLT-related topics in language teacher education courses to equip the teachers with requisite skills and expertise.
